Ac-Arg-Leu-Arg-AMC is a fluorogenic peptide that is usually the substrate of choice for assaying activity of purified proteasomes. Some substrates with R-AMC bonds can be used for assaying trypsin-like activity. Ac-Arg-Leu-Arg-AMC has a low Km, high specific activity, and is cleaved exclusively at the Arg-AMC bond.
Activity is quantified by release of free fluorescent 7-amino-4-methylcoumarin (AMC) which excites at 360-380 nm and emits at 440-460 nm.
